The Backup Exec Remote Agent debug log reports the following error while the job is running:
[2856] 05/16/10 09:00:19 VDiskMounter::MountAllDisks: failed to GetVolumeInformation, error 1392, \\?\C:\Program Files\Symantec\Backup Exec\Data\VMNAME-15-05-2010-12-05\MP00\E\
[2856] 05/16/10 09:00:21 VDiskMounter::MountAllDisks: failed to GetDiskFreeSpaceEx, error 1392, \\?\C:\Program Files\Symantec\Backup Exec\Data\VMNAME-15-05-2010-12-05\MP00\E\
VM-Ware support confirmed that this limitation is caused by vixMntapi not being able to access anything beyond the first 2 TB of a mounted volume.

The vixMntapi functions can be used to mount a Windows volume larger than 2TB. However, attempts to access files on the resulting mount may result in this error message: “The file system structure on the disk is corrupt and unusable. Please run the chkdsk utility on the volume.” Customers should not run chkdsk in this case. Even if the message does not appear, there is a hazard of corrupting the mounted volume if you attempt to change it, and a risk of getting incorrect data if the accessed file exists beyond the 2TB limit. There is no good workaround. Splitting the volume is not always possible or convenient.
